A nested case-control study within the LURIC cohort (3316 patients) applied single-cell multi-omics to 44 individuals with and without coronary artery disease (CAD) to identify immunological cardiovascular risk determinants. Multi-omics factor analysis uncovered a T-cell proliferation and interferon-signalling signature associated with CAD and cardiovascular mortality independently of hsCRP and IL-6, replicated across 136 independent scRNA sequencing datasets. These immune signatures may represent future tools for cardiovascular risk stratification.
