The SLC6A4 promoter tandem repeat polymorphism was examined in the SMOKING GENES case-control study (889 subjects) and the LURIC cohort (1,955 subjects). Genotype frequencies did not differ between smokers and non-smokers in either study (P = 0.13 and P = 0.33, respectively), and no association was observed with Fagerstrom score, pack-years, or daily cigarette count. These results indicate that the SLC6A4 promoter variant is not a major determinant of smoking behavior in Caucasians.
