The rs599839 A/G polymorphism near the sortilin 1 gene was examined for associations with LDL and triglyceride metabolism and cardiovascular risk in the LURIC cohort. G-allele carriers showed lower LDL cholesterol, LDL triglycerides, apolipoprotein B and triglycerides than AA homozygotes, and each G-allele reduced CAD prevalence (OR 0.806, 95% CI 0.692-0.940, P = 0.006). GG homozygotes had an OR of 0.588 for CAD and 0.693 for prior myocardial infarction, independent of cardiovascular risk factors.
