The influence of two TAFI polymorphisms – Thr325Ile and Ala147Thr – on cardiovascular and thrombotic events was examined in more than 3,300 LURIC patients. The Ile/Ile genotype at position 325 was associated with stroke incidence and earlier age at first stroke onset, with risk further elevated in patients with diabetes mellitus, myocardial infarction, or hypertension. No association was identified for Ala147Thr, pointing to a specific pathogenetic role for the Thr325Ile variant in stroke.
