TCF7L2 variants rs1225537 and rs7903146 were examined in 3185 LURIC participants, of whom 1021 had type 2 diabetes. Risk allele carriers showed earlier onset of type 2 diabetes, elevated HbA1c, reduced insulin secretion, and markedly increased proinsulin-to-insulin ratio (all p < 0.03), whereas GLP-1 concentrations were not significantly associated with TCF7L2 genotype. These findings extend to Caucasians the role of TCF7L2 in impaired proinsulin conversion and earlier diabetes onset.
