Among 2970 LURIC participants (median serum 25(OH)D 15.5 ng/mL), 960 SNPs across 29 skin-pigmentation genes were tested for association with 25(OH)D concentration; 46 reached p < 0.05. One EXOC2 variant survived false discovery rate correction with a delta 25(OH)D exceeding 5.00 ng/mL, and 11 SNPs in TYR, PRKACG, EDN1, TYRP1, and MITF also reached FDR significance. These findings indicate that variants in pigmentation-related genes predict vitamin D status in the Caucasian population.
