Seven common APOC3 variants were associated with circulating apoC-III in 3041 LURIC participants (all p < 0.05); alleles raising apoC-III also markedly elevated triglycerides and VLDL cholesterol but not LDL cholesterol or total apoB. In a meta-analysis of up to 332,389 participants, none of the variants was associated with coronary artery disease (all p > 0.1). These findings suggest that common APOC3-driven apoC-III elevation amplifies triglyceride-rich lipoprotein levels without increasing atherogenic burden.
