ApoC-II concentrations and cardiovascular mortality were analysed in 3,141 LURIC participants, 590 of whom died from cardiovascular disease over a median 9.9-year follow-up. An inverse J-shaped association emerged between ApoC-II quintiles and cardiovascular mortality; all quintiles above the lowest showed reduced risk after multivariate adjustment for ApoC-III (all p < 0.05). Enzymatic assays revealed a bell-shaped ApoC-II dose-response on GPIHBP1-LPL activity, suggesting optimal concentrations are required for maximal lipoprotein lipase function.
