Analysis of type 2 diabetes heterogeneity with a tree-like representation: insights from the prospective German Diabetes Study and the LURIC cohort

A tree-based dimensionality-reduction method stratified pathophysiological heterogeneity in 927 individuals with recent-onset type 2 diabetes from the German Diabetes Study, with 794 LURIC patients contributing validation for heart failure and mortality. The insulin-resistant branch had the highest hepatic lipid content, IL-6, IL-18, and cardiovascular risk, while the low-secretion branch carried the greatest neuropathy burden; in LURIC, all-cause mortality was highest in the insulin-resistant branch (pdim2 = 0.0032).
